  •        核仁小rna (snorna)是一类中等长度的非编码小rna,其长度60-400nt不等,与核仁核糖核蛋白结合形成snornps复合物发挥功能[1]。在脊椎动物中,核仁小rna的编码基因主要存在于蛋白编码基因的内含子区,经转录加工处理形成成熟的核仁小rna分子[2]。snorna参与多种生物学过程,比如rrna的加工处理,rna剪接和翻译,以及氧化应激反应的调控[3]。snorna主要通过snornps复合物来发挥作用。通过snorna碱基互补识别靶基因特定位点,snornps复合物催化这些位点的2’-o甲基化或假尿嘧啶化反应。根据snorna结构与功能的不同,可以将snorna分为两大类:负责2’-o甲基化的box c/d snorna和负责假尿嘧啶化的box h/aca snorna [1]。另外,还存在一类比较特殊的snorna- scarnas (它们具有类似box c、box d或者box h、box aca结构,特异表达于small cajal body) [4]。snorna可以产生更小片段的非编码rna-sdrna。sdrna与mirna类似,能够通过调节mrna的稳定性来发挥作用 [5]。

           多项研究表明,snorna在肿瘤中异常表达,并在肿瘤的转移过程中扮演着重要角色 [6-7]。在乳腺癌和前列腺癌中,snord50a发生了2个碱基的缺失突变,并且其拷贝数明显降低,因此被认为是抑癌基因[8]。有趣的是,h/aca box snorna snora42在肺癌中作为癌基因存在,抑制snora42的表达具有明显的抗癌作[10]用。c/d box snornas的表达水平常伴随肿瘤的发生而增加 [11]。有一些c/d box snornas可以作为疾病的分子标志物,比如snord44和snord43与乳腺癌的不良预后相关[9]。snorna在神经退行性疾病的发生发展过程也发挥着关键作用。snornas在血液、痰液以及尿液中稳定存在,使其作为疾病分子诊断标志物方面具有巨大的前景。目前,snorna已经成为疾病领域特别是癌症领域里的研究热点。
           nrstar™ human snorna pcr芯片包括359条snorna、7条snorna靶向snrna以及4条snornp复合物成员mrna,而nrstar™ mouse snorna pcr芯片分别包含370个snorna。该pcr芯片覆盖了所有pcr技术可区分的snorna,能够快速方便的检测出组织、细胞以及血液中的snorna的表达情况。它还可应用于疾病分子标志物的高通量筛选和验证。
           为了确保数据质量,nrstar™ human snorna pcr芯片包含3个针对人snorna的内参基因和2个针对人snornp复合物成员mrna的内参基因,而nrstar™ mouse snorna pcr芯片包含8个针对小鼠snorna的内参基因用于数据的均一化处理。此芯片还使用rna spike-in,pcc和gdc对照组来检测cdna合成效率,pcr扩增效率和基因组dna污染。

           为保证nrstar™ snorna pcr array(h/m)的实验效果,建议配套使用arraystar rtstar™ first-strand cdna synthesis kit (as-fs-001)
